3 VERITY 3011 Isocratic Pump: Consistent and Reproducible Pump Performance TECHNICAL NOTE TN211 GILSON APPLICATIONS LABORATORIES INTRODUCTION The VERITY 3011 Isocratic Pump offers accurate flow rates and virtually pulse-free flow, making it ideal for applications in the chemical, pharmaceutical, environmental, food and beverage, biotechnology, and petroleum industries. Two stainless steel pump heads are available, with maximum flow rates of 5 ml/min or 10 ml/min and a maximum pressure of 600 bar, allowing the pump to be used for analytical to semi-preparative applications. Pump heads can be easily and rapidly changed using a quick-connect system without the use of tools or software control. The VERITY 3011 Pump functions as a stand-alone instrument using its own embedded software and can be incorporated into a modular system such as high pressure liquid chromatography (HPLC) or gel permeation chromatography (GPC). Three modes of operation are available on the VERITY 3011 Pump embedded software: Flow mode (delivery of a constant flow rate), Dispense mode (delivery of a specified volume or pump for a specified time), and Program mode (executes a predefined method). This technical note presents data collected in Flow mode and Dispense mode, demonstrating that the pump can deliver consistent and repeatable flow rates over time (Flow mode), and can provide consistent and repeatable large volume injection onto an HPLC system (Dispense mode). Refer to TN 214 (VERITY 3011 Isocratic HPLC Separation of common Pharmaceutical Compounds) for details on using Program mode. MATERIALS AND METHODS Procedure Figure 1 VERITY 3011 Isocratic Pump Test A The VERITY 3011 Isocratic Pump, equipped with a 5 SS pump head, was operated inside a fume hood and controlled using its embedded software.the pump was set to Flow mode, and a mixture of 50:50 (v/v) ethanol:water pumped at 1 ml/min and approximately 17.2 bar (250 psi). The pump was configured via the Settings option to use the fluid Other Liquid with a compressibility of 79 Mbar -1 to optimize the pumping for the ethanol:water mixture. During the four-hour test, 5 ml samples were obtained every 15 minutes. Each of these collections involved 10 minutes of not collecting volume, followed by 5 minutes of collecting volume. 4 Test B The VERITY 3011 Isocratic Pump with a 10 SS head was used as an injection pump to load sample directly onto an HPLC column. The modular Gilson HPLC system was comprised of a 322 Pump, 159 UV/VIS Detector, and 506C System Interface. The VERITY 3011 Pump and 322 Pump were plumbed to the column via a tee. TRILUTION LC v3.0 Service Pack 8 (SP8) was used to control all system components except for the VERITY 3011 Pump, which was controlled by its embedded software. Communication between the two software applications was accomplished by contact coordination between the 506C System Interface and the VERITY 3011 Pump. Figure 3 VERITY 3011 Pump Software in Dispense Mode The HPLC system was operated at 20 ml/min using 25:75 methanol:water (isocratic) as the mobile phase with a 21.2 mm ID column. Ten injections of a 0.1 mg/ml caffeine in water solution were performed. A TRILUTION LC SP 8 sample list consisting of ten methods was created. The VERITY 3011 Pump was configured for the appropriate head size and solvent compressibility, and the run mode set to dispense 10 ml at a flow rate of 5 ml/min. Software and Hardware Test A VERITY 3011 Pump (5 SS head) Graduated Cylinder (Pyrex 3022) Test B TRILUTION LC v3.0 Service Pack Pump (H2 head) 159 UV/VIS Detector (0.5 mm flow cell) VERITY 3011 Pump (10 SS head) Phenomenex Luna, 50 x 21.2 mm, 5µm column (Product Code 00B-4252-P0-AX) RESULTS AND DISCUSSION Test A The pump was operated in Flow Mode at a constant rate (1 ml/min) dispensing a solution of 50:50 (v/v) ethanol:water for four hours in cycles of 10 minutes of no liquid collection, followed by 5 minutes of liquid collection. As shown in Table 1, the volume measured in each collection period was consistent and with minimal variation across samples. This demonstrates that the pump can deliver consistent and accurate flow rates. 5 Test B The TRILUTION LC SP 8 sample list ran until completion. For each method iteration, the VERITY 3011 Pump was started in Dispense mode at 5 ml/min via an input contact signaled from the HPLC system. Flow stopped after the VERITY 3011 Pump dispensed the requested volume of 10 ml onto the column. Chromatographic data from the ten injections overlaid can be seen in Figure 4, and the CV for peak area of the ten injections is presented in Table 2. The VERITY 3011 pump can produce consistent dispense volumes as shown by the chromatogram overlay and quantitatively with the peak area CV% calculation. 7 VERITY 3011 Pump: Isocratic HPLC Separation of Common Pharmaceutical Compounds TECHNICAL NOTE TN214 GILSON APPLICATIONS LABORATORIES INTRODUCTION The VERITY 3011 Isocratic Pump features a virtually pulse-free, stable solvent flow rate, ensuring stable flow delivery to detectors, and allowing for a wider range of application capabilities on one pump. The pump also features an easily interchangeable quick-connect pump head, making maintenance simple and reducing lengthy downtime. As a stand-alone pumping system with integrated touchscreen, the VERITY 3011 Pump is ideal for enabling reaction monitoring, and supporting both analytical and semi-preparative chromatography systems for environmental, food and beverage, and petroleum applications. The pump allows for flow rates up to 10 ml/min, and pressures of up to 600 bar. This technical note highlights the ability of the VERITY 3011 Pump to perform an isocratic HPLC separation of a solution containing 4-acetamidophenol, theophylline, and caffeine. During the first test, the pump was operated by TRILUTION LC v3.0 with Service Pack 8 (SP8), whereas the pump was programed with its own embedded software during the second test. Figure 1 VERITY 3011 Pump with Solvent Tray and Column Holder MATERIALS AND METHODS Procedure Test A: The VERITY 3011 Pump was integrated into a Gilson HPLC system equipped with a GX-271 Liquid Handler, 157 UV-VIS Detector, and operated by TRILUTION LC v3.0 with Service Pack 8. The degassed 25:75 methanol:water isocratic mobile phase was run at 1 ml/min with a 4.6 mm ID column. Ten total loop injections were performed with a solution of 0.1 mg/ml 4-acetamidophenol, theophylline, and caffeine in 50:50 methanol:water. 8 Test B: The same procedure was performed as in Test A; however, the VERITY 3011 Pump was controlled using its embedded software. The other instruments the liquid handler and detector remained under the control of TRILUTION LC software. Communication between the two software applications was accomplished by contact coordination between the GX-271 Liquid Handler and the VERITY 3011 Pump. This configuration is analogous to using the VERITY 3011 Pump with non-gilson software. Two VERITY 3011 pump methods were created using the embedded software. These included a method to run the pump and coordinate with TRILUTION LC, and a method to stop the pump at the end of the run. This second method was set as the Linked Method in the first, so that the pump would automatically shut down upon completion. The pump was then configured for the appropriate head size and solvent compressibility, set to Program Mode, and the method set to loop ten times. The three compounds were separated for each of the ten injections. Chromatographic data from the ten injections overlaid can be seen in Figure 5, and CVs for the retention time and area of the ten injections are presented in Table 1. The pressure data from the VERITY 3011 Pump was collected during each run, and was steady over the course of each injection. No pressure fluctuations are observed in the pressure following appropriate equilibration, indicating a consistent flow from run-to-run. Figure 6 Test A, Injection 1, with Pressure Trace Test B The TRILUTION LC sample list ran until completion, and contact communication between TRILUTION LC and the VERITY 3011 Pump s embedded software started the pump, ran the pump method for ten loops, and then stopped the pump. TRILUTION LC collected detector data for each of the ten injections. The three compounds were separated for each of the ten injections. Chromatographic data from the ten injections overlaid can be seen in Figure 7 and CVs for the retention time and area of the ten injections are presented in Table 2. This test demonstrates that the VERITY 3011 Pump can be effectively controlled via non-gilson software using contact coordination, and a comparison of the results from the ten injections show that the pump can provide a consistent flow rate.
