3 CALIBRATION The items you need for calibration are: Fluorometer Standard Syringe 2.5 ml Black cuvette (calibration blank) Cuvette 1. Make sure all chemicals are equilibrated to room temperature Turn on the fluorometer Press on/off Display should read UV and 0.0 3

4 CALIBRATION 2. Press STD VAL on the fluorometer Make sure the value is the same as listed on the reverse side of the fluorometer 3. Press CAL 4. Press ENTER 5. At the prompt, place the black cuvette in the fluorometer and press ENTER Note; if you do not push enter right away after pushing cal, the instrument will return to standby mode. If that happens simply press cal again to restart the process. 4

5 CALIBRATION 6. Attach the syringe to the standard bottle 7. Push the syringe and the syringe mounting platform all the way down. There is a click 8. Turn the standard bottle up side down. Transfer 2.5 ml standard to the syringe Remove air bubbles in the syringe by pumping up and down 5

6 CALIBRATION 9. Transfer the standard to the cuvette 10. Remove air bubbles by gently tapping the cuvette with a finger nail 6

7 CALIBRATION 11. Place the cuvette in the fluorometer. Close the lid and press ENTER 12. Press ENTER to finalize the calibration 13. Leave the cuvette in the fluorometer and press READ The value should be within 3 % of the Standard value This value is termed Standard Value Measured 14. Note the Standard Value Measured on the analysis sheet or directly on the Excell spreadsheet You have now calibrated the fluorometer 7

8 Analytical Blank 1. Transfer developer to a cuvette The dispenser is operated correctly by pushing the plunger all the way down in one slow and steady movement Whenever a dispenser is taken into operation, discard the first pumping in order to remove air bubbles For more information on operating the dispenser please refer to Dispenser Notes 2. Place the cuvette with the developer in the outermost right square on the rack 8

9 Analytical Blank 3. Attach a 1 ml syringe to the substrate bottle 4. Push the syringe and the syringe mounting platform all the down. There is a click 5. Turn the substrate bottle up side down. Transfer 0.35 ml substrate to the syringe Remove air bubbles in the syringe by pumping up and down 9

10 Analytical Blank 6. Transfer the substrate to the developer in the cuvette Mix by pumping the liquid up and down three times in the cuvette using the syringe. Remove air bubbles in the cuvette 7. Place the cuvette in the fluorometer and close the lid 8. Press READ This value is termed the Blank Value 10

12 Sample FILTRATION 1. Note date, sample ID and room temperature (recommended C) in the analysis sheet or directly in the spreadsheet 2. Note the sample ID on the filter 12

13 Sample FILTRATION 3. Transfer water sample to the sampling syringe and note the sampling volume in the analysis sheet 4. Attach the sampling syringe to the filter 13

14 Sample FILTRATION 5. Push the water sample through the filter and into an appropriate waste bottle 6. Evacuate the dead volume in the filter by detaching the syringe from the filter. Fill it with air, reattach the syringe and use the air to push out the remaining liquid in the filter into the waste bottle 14

15 ANALYSIS 1. Retrieve substrate bottle, filtered samples, blunt needles, cuvettes, glass waste vial and 2.5 ml syringes, according to the number of samples to be analyzed 2. Transfer 2.5 ml substrate to each syringe Remove air bubbles in the syringe by pumping up and down 15

16 Analysis 3. Gently attach the syringes on the filters. Then place the syringes with filters on the rack as shown in the picture 4. 17 Analysis 5. Flush the filter with substrate 6. Surplus liquid is collected in the glass vial for disposal 7. Attach the filter with syringe to the blunt needle 8. Rapidly repeat the routine for each sample 9. Start the stopwatch 17

18 ANALYSIS 10. After 15 minutes the assay is terminated by placing the filter with the syringe and blunt needle in the developer and back flushing two times 11. Turn on the fluorometer 12. Gently remove air bubbles in the cuvette 13. Place the cuvette in the fluorometer. Close the lid 14. Press READ 18

19 ANALYSIS 15. Note the fluorescence reading (analysis value) on the analysis sheet or directly on the spreadsheet 16. Use the spreadsheet to calculate the BQ value 19

20 DISPENSER NOTES BEFORE USE 1. Make sure the dispenser is attached correctly to the bottle. 2. Make sure that the developer dispenser is correctly set (2 ml) before use. 3. Once the dispenser is mounted on the bottle it is important to keep it in an upright position to prevent leakage. 4. Remove trapped air inside the dispenser, by dispensing 3 times and discard the fluid. Dispenser is now ready for use. 20

21 DISPENSER NOTES OPERATION 1. The dispenser is operated correctly by pushing the plunger all the way down in one slow and steady movement. 2. Whenever the dispenser is taken into operation, check setting (2 ml), push the plunger down once and discard the dispensed developer in order to remove air bubbles. MAINTENANCE 1. Prior to changing developer bottles it is necessary to rinse the interior of the dispenser thoroughly with distilled water (not necessary the first time), by repeated dispensing. Remove water inside the dispenser before refitting. 2. For long time storage, flush the dispenser with distilled water as described above. 21
