Be advised that the VP 710C1-M Magnetic Tumble Stirrer has very strong magnetic fields coming from a 48 MGO Neodymium Iron Boron magnetic cylinder. People with pacemakers should not get closer than 24 inches to the Tumble Stirrer. Remove all magnetic field influenced tools and objects from the immediate area to prevent them from being pulled onto the magnet or from striking anyone as the objects are pulled towards the magnet. Unless a non-magnetic screwdriver is available, use caution when using a screwdriver to remove and attach screws to the VP 710C1-M Tumble Stirrer. It will be pulled towards the magnet. Keep credit cards, watches and other magnetic sensitive items at least 24 inches from the VP 710C1-M Tumble Stirrer s magnetic fields. 3 INTRODUCTION The VP 710C1-M-Kit includes the VP 710C1-M Magnetic Tumble Stirrer, VP 750PTFE Bubble Paddle Reservoir, FXALP-PMS (a modified FX ALP with a pocket mount feature), removable Adaptor Plates and Leveling Foot (see parts photo, page 2). The VP 710C1-M Magnetic Tumble Stirrer (1) has modified features which allow it to be set up on various automated workstations by attaching it to a modified ALP plate holder or attaching different adaptor plates to the bottom of the VP 710C1-M. The VP 710C1-M can be secured to the underside of the FXALP-PMS (4) for use on the Beckman Biomek FX or NX workstations. With the VP 581-PE Adaptor Plate (5) attached to the underside of the VP 710C1-M, the two locating pins in the VP 581-PE can be aligned to locating holes on a PerkinElmer EP3 or Janus deck. With the VP 581B-2 SBS Adaptor Plate (6) attached, it can be placed in a plate holder/carrier on a Tecan, Hamilton or other workstation decks with SBS plate holders. The VP 750PTFE Bubble Paddle Reservoir (9) is a stirring reservoir used to keep beads, cells or particulates in suspension during automated pipetting operations (must be ordered separately from VP 710C1-M-Kit). It has a VP 581B SBS Adaptor Plate attached to the bottom of the reservoir. This allows the reservoir to fit in the Plate Holder Template (8) on the VP 710C1-M stir deck or in the pocket mount feature of the FXALP-PMS (4). The Bubble Paddles inside the reservoir are driven by the magnetic field from the VP 710C1-M Magnetic Tumble Stirrer. (NOTE: To avoid splashing do not let the reagent level in the reservoir go lower than the top of the bubble paddle.) The Leveling Foot (7) is used to support the motor of the VP 710C1-M when mounted on each workstation, keeping it level during operation. 6 SET UP The VP 710C1-M-Kit contains a multi-use model of the popular VP 710C1 Magnetic Tumble Stirrer along with the VP 750PTFE Bubble Paddle Reservoir, which is used to keep beads, cells and particulates in suspension during pipetting operations. The Kit is designed for operation on many common automated workstations. Instructions for use on the Beckman, PerkinElmer, Tecan and Hamilton systems are detailed below. Set Up For Biomek FX And NX Workstations: Using the provided 3/32 Allen wrench, remove the four set-screw plugs from the FXLP-PM attachment holes in positions 1, 2, 9, and 10 (circled in photo below). Store them in the labeled plastic bag Attach the Leveling Foot (7) on narrow side of VP 710C1-M: Select an appropriate length of threaded rod from the various sizes provided. Place the tightening nut, then the washer, on the rod selected (see photo below). Carefully screw the Leveling Foot into the threaded hole on the narrow side of the VP 710C1-M. Note: The most common threaded rod length for the FXALP-PMS set up is 6.3 cm. Nut Washer Do not tighten the nut until the VP 710C1-M is placed on the workstation. 7 Turn the VP 710C1-M over so the Leveling Foot is under the motor. Slide VP 710C1-M deck under the top plate of the FXALP-PMS (4) through the narrow side of the ALP. NOTE: A stack of two microplates may be placed under the VP 710C1-M for support while aligning the appropriate attachment holes and inserting the screws. Attach the top plate of the ALP to the narrow side of the VP 710C1-M deck with four flat head screws (10, circled in photo below). Remove microplate supports. Microplates Secure the bottom plate of the FXALP-PMS with attached VP 710C1-M to the deck of a Biomek FX or NX using the thumb screws (see photo below). The length of the VP 710C1-M should be parallel to the front edge of the FX or NX deck. Leveling Foot Thumb Screw V&P Scientific, Inc. 7 Technical Note 186
8 Adjust the Leveling Foot so the VP 710C1- M motor and deck are level. Move the tightening nut until it is securely against the washer at the bottom of the VP 710C1-M. This will lock the Leveling Foot in place 1 3 Attach the internal power cord between the VP 710C1-M and the Power Supply (white arrows). Attach the external power cord to the power supply (red arrows). Plug into appropriate power outlet Place the VP 581B SBS Adaptor Plate with the VP 750PTFE Bubble Paddle Reservoir into the pocket feature of the FXALP-PMS. Tighten the four thumb screws to secure the VP 581B SBS Adaptor Plate to the ALP. Confirm X, Y and Z positions of the pipette tips to the bottom of the reservoir. Thumbscrews Turn on the power supply. Adjust the speed of the Bubble Paddles to a moderate speed. With the paddles spinning, pour the reagent to be stirred into the VP 750PTFE Bubble Paddle Reservoir. Begin pipetting operations. 9 NOTE: If the deck of the FX/NX is crowded with many accessories, it may be necessary to slide the VP 710C1-M under the FXALP-PMS, then lift the VP 710C1-M and FXALP-PMS together (but not attached to one another) onto the FX/NX deck over the selected ALP position. Slide the motor of the VP 710C1-M under an adjacent ALP, and then proceed with attaching the VP 710C1-M to the underside of the FXALP-PMS as described above. Alternative Set Up For Biomek Fx And Nx Workstations For Use With Optional VP Channel Bubble Paddle Reservoir (Not In Kit): Remove Plate Holder Template (8) and VP 581-PE (5), if attached, from VP 710C1-M Tumble Stirrer (as described on pages 11 and 12). Using the provided 3/32 Allen wrench, remove the four set-screw plugs from the FXLP-PM attachment holes in positions 1, 2, 5, and 6 (circled in photo below). Store the in labeled plastic bag Attach Leveling Foot to Tumble Stirrer on narrow side of the VP 710C1-M as described on page 6. Turn the VP 710C1-M over so the Leveling Foot is under the motor. Slide VP 710C1-M deck under the top plate of the FXALP-PMS through the wide side of the ALP. Microplates may be placed under the Tumble Stirrer for support, as described on page 7. Attach the top plate of the ALP to the narrow side of the VP 710C1-M deck with four flat head screws (10, circled in photo below). Remove microplate supports, if used. 10 Secure the bottom plate of the FXALP-PMS with attached VP 710C1-M to the deck of a Biomek FX or NX using the thumbscrews (red arrow to the left). The length of the VP 710C1-M will be perpendicular to the front edge of the FX or NX deck. Adjust the Leveling Foot so the motor and deck are level. Attach the power cords as described on page 8. Place the VP 581B SBS Adaptor Plate with the attached VP Channel Bubble Paddle Reservoir (not included in kit) into the pocket feature of the FXALP- PMS. Tighten the four thumb screws (white arrows in photo below) to secure the VP 581B SBS Adaptor Plate to the ALP. Confirm X, Y and Z positions of the pipette tips to the bottom of the reservoir. Turn on the power supply. Adjust the speed of the Bubble Paddles to a moderate speed. With the paddles spinning, pour the reagent to be stirred into the VP 750PTFE Bubble Paddle Reservoir. Begin pipetting operations. NOTE: To avoid splashing do not let the reagent level in the reservoir go lower than the top of the bubble paddle. V&P Scientific, Inc. 10 Technical Note 186
11 Set Up For Perkinelmer EP3 And Janus Workstations: Attach Plate Holder Template (8) to the deck of VP 710C1-M Tumble Stirrer as follows: Remove the four screws from the Plate Holder Template attachment holes (circled). Align the Plate Holder Template (8) to the four attachment holes on the deck of VP 710C1-M Tumble Stirrer. Replace the four screws into the same Plate Holder Template attachment holes. Turn the VP 710C1-M upside down. Remove the four Adaptor Plate attachment screws from the bottom of the VP 710C1-M. V&P Scientific, Inc. 11 Technical Note 186
12 Align the VP 581-PE PerkinElmer Adaptor Plate (5) to the end of the VP 710C1-M deck. Attach the VP 581-PE with the four attachment screws (see white arrows in photo below). Turn the VP 710C1-M back to the upright position. Align locating pins (red arrows in photo above) on the bottom of the VP 581-PE to locating holes on the deck of the EP3 or Janus. Attach the power cords as described on page 8. Place the VP 750PTFE Bubble Paddle Reservoir (9) in Plate Holder Template (8) on top of the VP710C1-M. The Leveling Foot is not needed with this configuration. Confirm X, Y and Z positions of the pipette tips to the bottom of the reservoir. Turn on the power supply. Adjust the speed of the Bubble Paddles to a moderate speed. With the paddles spinning, pour the reagent to be stirred into the VP 750PTFE Bubble Paddle Reservoir. Begin pipetting operations. 13 Set Up For Tecan Freedom Genesis Workstations: Attach Plate Holder Template (8) to the deck of VP 710C1-M Tumble Stirrer using Plate Holder Template Screws as described on page 11. Turn VP 710C1-M upside down. Remove four Adaptor Plate attachment screws from bottom of the VP 710C1-M (Adaptor Plate attachment holes circled below). Attach VP 581B-2 SBS Adaptor Plate (6) to bottom of VP 710C1-M with the Adaptor Plate attachment screws. V&P Scientific, Inc. 13 Technical Note 186
14 Adjust the Leveling Foot to nearly the appropriate length by taking apart or connecting threaded rod segments to lengthen or shorten. Attach the Leveling Foot into the threaded hole on bottom of VP 710C1-M. Turn the VP 710C1-M back to upright position (Leveling Foot down). Place the VP 710C1-M on the deck of a workstation using the VP 581B-2 SBS Adaptor Plate aligned in a plate holder. Tecan Plate Carrier Adjust the Leveling Foot so the VP 710C1-M motor and deck are level. Move the tightening nut until it is securely against the washer at the bottom of the VP 710C1-M (see photo left). This will lock the Leveling Foot in place. Attach the power cords as described on page 8. Place the VP 750PTFE Bubble Paddle Reservoir (9) in Plate Holder Template (8) on top of the VP710C1-M. Confirm X, Y and Z positions of the pipette tips to the bottom of the reservoir. Turn on the power supply. Adjust the speed of the Bubble Paddles to a moderate speed. With the paddles spinning, pour the reagent to be stirred into the VP 750PTFE Bubble Paddle Reservoir. Begin pipetting operations. 16 OPERATION We recommend that the Tumble Stirrer is installed as far away from ferromagnetic material as possible. The closer and larger the magnetic material is to the Tumble Stirrer, the greater the torque that is required and the slower the maximum speed will be. Also, although aluminum is not ferromagnetic it will cause a drag on the magnetic field because eddy currents are created in the aluminum when magnetic lines of flux pass through it. A larger mass of aluminum will cause a greater drag and more heat will be generated by the eddy current. OPERATING SPEED The operating speed of the Tumble Stirrer is application dependent and needs to be determined empirically. The Maximum speed set at the factory is 2150 RPM when the knob is set to 100 and there is no load. Because the load (magnetic resistance) will directly affect speed, it is not possible to accurately relate the dial setting to speed (RPM) in every situation. In most cases, the knob has to be set to at least 20 to overcome inertia. If the Tumble Stirrer is placed near a ferromagnetic object, a higher dial setting is required to overcome inertia. Once the optimal stirring speed has been determined, note the position and leave the speed knob set to that position. Factors to consider in determining optimal stir speed are the fragility of the objects being stirred, size, shape and depth of the microwells, composition of the microplate (polypropylene or polystyrene), volume and viscosity of the liquid and the type of Stir Disc, Stir StiX or Stir Bar used (such as whether they are stainless steel, Alnico, Samarium Cobalt, or Neodymium Iron Boron) as well as the shape of the Stir Element. To stir very viscous solutions, start at a low speed, and then ramp up slowly to the desired RPM. The actual RPM can be determined with the VP 710-RPM Hall Sensor RPM Meter which can be purchased from V&P Scientific as an additional component of the system. In general, stirring microbial cultures works best at low speeds. Stirring to re-solubilize extracts or to stir in deep well microplates requires high speed. Again, the speed of stirring needs to be determined empirically for each particular application. The control unit for the Tumble Stirrer has an ON/OFF power switch and a speed control knob. Place the power switch in the ON position (toggle up), then adjust the speed control knob to change the operating speed of the Tumble Stirrer. The speed control for the Tumble Stirrer is designed to control the speed and to gradually take the unit from the OFF position to the set speed in a gradual ramp up. The speed control knob should not be used to stop the motion of the Tumble Stirrers. To stop the Tumble Stirrer, always flip the power switch to the OFF position (toggle down). 17 CARE When not in use, turn the power switch off. Do not place the control unit in chambers with temperatures above 40 o C. The case and deck of the Tumble Stirrer is made of ABS. To clean, use a mild detergent followed by a water rinse. For chemical compatibility of ABS please see the following link for more information: The VP 710C1 series Stirrers are powered by a powerful, brush-type DC servo motor. Occasionally the brushes need to be changed. Extra fuses (10A, 250V fast, input fuses and 5A, 250V slow, output fuses) have been provided in the rare event that they blow. The extra fuses are inside the control unit. Fuse Replacement The VP 710C1 series Stirrer is powered by a Multi-Drive Solid State DC Motor speed controller, which is manufactured by KB Electronics, Inc. The controller is powered by 115/230 VAC at 50/60 Hz. Please refer to the KB Control Installation and Operating Instructions (attached) before operating the controller. The controller contains two fuses to protect against over-current. The first one is an AC in-line fuse for the VP710C1 series controller which is located on the right rear of the circuit-board (see Page 5 of the KB Controller operating instructions). The AC in-line fuse should be a FAST- ACTING 250V ABC 10A fuse. The second one is a DC output fuse on the KB Controller, which is located on the left rear of the circuit board (see page 5 &13 of the KB Controller operating instructions). The DC out-line fuse should be a SLOW-BLO 250V MDL 5A fuse. A set of spare fuses is located inside a bag taped to the inside top cover of the controller. 18 SPECIFICATIONS AND CAUTIONS If the equipment is used in a manner not specified by V&P SCIENTIFIC, INC., the protection provided by the equipment may be impaired. The use of motor controls, like that of all utilization of concentrated power, is potentially hazardous. The degree of hazard can be greatly reduced by proper design, selection, installation, and use, but all hazards cannot be completely eliminated. The following safety precautions must be observed during all phases of installation, operation, service, and repair of this motor control product. Failure to comply with these precautions or with specific warnings elsewhere in this manual violates safety standards of design, manufacture and intended use of the products. V&P Scientific assumes no liability for the customer s failure to comply with safety requirements and practices. Note the following WARNINGS and CAUTIONS associated with the Tumble Stirrer. A WARNING will highlight a procedure, which presents potential danger to people, and a CAUTION will highlight a possible danger to equipment. Both types of instructions must always be followed. WARNING Do not touch the amplifier or components on the PCB right after turning off the power. Wait until the power indicator turns off or at least seven minutes. Do not attempt to wire circuitry while power is on. Do not attempt to examine the components and signals on the PCB while operating. Do not attempt to disassemble or modify internal circuitry, wiring, or components. The grounding terminal must be grounded properly with 200V class type III standard. Do not remove the cover of the motor case when the power is ON to avoid personnel injury caused by electrical shock. CAUTION Do not attempt a dielectric strength test to internal components. They are sensitive semiconductor-devices vulnerable to high voltages. Do not connect the output terminals to an AC power outlet. Do not disconnect motor during operation.
