5 The biomedical division of the Linari Engineering srl company is an international reference for the production of electrospinning advanced systems and accessories and now it is also expanding its innovative systems offer for the in vitro cell culture. Investing in electrospinning is the most economic and promising way to discover the world of micro and nanofabrication with a wide variety of materials and incredible applications possibilities. Starting from a material dissolved in a solvent, one can create continuous filaments of diameters ranging from a few tens of nanometers up to several micrometers, simply by varying the high voltage level or the concentration of the solution itself. That is to say that mechanical operations on sophisticated micrometric systems are not necessary to enter the nano scale world. 11 Electrospin theory Tubolar structure made with a roto-translating collector of few millimeters in diameter. Suitable for tissue engineering and fi ltration thanks to uniform thickness. Having a simple electrospinning setup, made from an high voltage generator and a syringe pump is possible to create high quality substrate over rotary and translating collectors connected to RT Collector WEB. Small and lightwei-ght system can be placed inside the electrospinning chamber to create tubolar structures with random and even aligned nanofibers. With a translational movement up to 200 mm it s able to obtain uniform thickness along all the collecting drum. A dielectric structure allows the high voltage connection to the collector up to +/- 40 kv without any risk for motors and drivers. 18 Linari Engineering

21 RT COLLECTOR The RT Collector WEB can be connected, with proper options features, to any analog high voltage generator thanks to a fail safe interlock for chamber access. Up to two generators can be driven at the same time from the 5 LCD display and PC software (option). The direct high voltage control from RT Collector control system allows: Single start/stop for movement and HV generators, Voltage level coordination with movement speed and position, Digital interface for voltage and current intensity control, Optocoupled analog signals to avoid damage during accidental sparking. All collectors (disks and drums with lenght lower than 150 mm) can be installed in a minute thanks to fast grips. For disks of larger diameter the support arc must be dismounted. 37 PUMP Single syringe pump The Model BSP- 99M can accommodate from a microliter syringe up to a 50cc syringe, both in plastic and in glass. The Model BSP-99M has a built-in fl ow chart, calibrated for the Multifi t with 99 equally spaced speeds. Its simple and strong construction makes this pump a good solution for electrospinning setup even with high voltage directly connected to the needle. 43 coaxial needles Modular version Coaxial needles are suitable for core-shell electrospinning with two or three different coaxial solutions. The modular execution of these needles allows the simple installation of tips over each different layer. Different diameters for each layer are available in stock as shown in the following table. Custom connector from needle and base was designed to warranty high repetibility during usual dismounting and handling cleaning procedure. Having the triple layer version is always possible to spin with just two layers simply dismounting the last, external layer with just few screws. Each layer is supplied with an independent fl uid inlet placed on the rear side where can be placed a LUERUNF adaptor to obtain a standard Luer Female connector to connect it to a syringe with tefl on or PVC fl exible pipes. The coaxial needle symmetrical shape and smooth surface assure a good quality electric fi eld to obtain a stable nanofi bers prodution. All our needles are made of AISI 316 stainless steel and sealings are made of NBR rubber o-rings. Other materials available on request. 93 CELL COLTURE 3D embrion The new frontiers of perfusion bioreactors is the integration with microchannel structures to create a well known three dimensional fl ux of chemicals and nutrients. Using a monolithic disposable mould made of FDA approved silicone, we create a complete infusion system able to generate precise fl ow mixture of different fl uids (up to eight different ones) to obtain a 3D concentration gradient. Inside the mould there is a cubic inch free volume where can be placed the scaffold. The fl uid mixing is done by an accurate microchannel structure engraved on four faces of the silicone mould. The sample is well observable from two ortogonal windows under microscope or other optical probe. 3D Embrion is not just an advanced bioreactor specially designed for colture of cells, but it can be used to create special hydrogel matrix with 3D gradient of mechanical properties. It can have several applications for the study of phenomena related to cell mechanotaxis (cells tend to migrate from soft regions to more rigid ones when they are seeded on a substrate with a gradient of stiffness), or differentiation on different stiff substrates. 95 Perfusion bioreactor Rotary wall perfusion bioreactors Our rotary wall bioreactor join a simple and essential design with a 15 x 25 cm footprint able to locate up to two colture chambers, even of different volume. Both are moved from the same motor with a gear to warranty a continuos and regolar rotation even after long operation time. The control system drive the motor with high speed accuracy from 0.1 up to 50 rpm with a simple switch from continuos rotation and shaking mode with alternative rotation clockwise and counterclockwise. The controller can integrate up to two peristaltic pump in the same case. The rotary chamber is simply placed in its housing with a vertical movement without any lock. The fl uid inlet and outlet are done from the central pipe. Thanks to radial holes on the two ends we had an uniform nutrient distribution. Chamber diameter is 60 mm but several lenghts are available. Mixed fluxes perfusion bioreactors new concept for A the classic cellular bioreactor allows to place sponge and fl at scaffold in the same case. This reactor has eight fl uid connections: four over the scaffold and the other ones below it. If the scaffold is placed over a grid in the middle of the height it is possible to provide different environment on the two faces of the scaffold. It can be useful for epitelial cells where the lower fl uid can be used even to create mechanical stress with a pressure pulse generator. The high accuracy shape of the upper fl uid inlet helps creating a fl at fl ow rate profi le if the system is used as a standard perfusion bioreactor with fl uid outlet from the bottom connections. High fl uid connection availability allows simple installation of instruments and sampling system directly inside the bioreactor. Internal diameter is 35 mm and the scaffold supporting grid can be provided in different mesh or as a sealed disk (see lower disk with o-ring in the picture). Biomedical Division 95
