1 Using Plates and Vials with ACQUITY UPLC and ACQUITY UPLC H-Class Systems Follow the guidelines in this document when using plates and vials with ACQUITY UPLC and ACQUITY UPLC H-Class systems. 2 Plates Follow these recommendations when selecting plates for use with the ACQUITY UPLC Sample Manager and Sample Organizer and the ACQUITY UPLC H-Class Sample Manager-FTN: ACQUITY H-Class In Empower and MassLynx, ACQUITY UPLC-compatible plate formats are identified by the ANSI designation, which indicates the plate meets the standards adopted under Society for Biomolecular Screening (SBS) guidelines. If you use plates from a manufacturer other than Waters, ensure that the plate size conforms to Waters' specifications by direct measurement or comparison of the manufacturer s specifications. Doing so is particularly important when using 384-well plates with smaller well sizes. Do not centrifuge plates. The plate can warp, which can cause an alignment fault. During testing, plates filled with 100% organic solvent (methanol, 2-propanol, or acetonitrile) or 50:50 mixtures of organic solvent and water, covered with polypropylene cap mats and stored in the Sample Organizer at between 10 and 15 ºC, ran successfully. At temperatures closer to ambient, plates filled with solutions containing high concentrations of organic solvents give less consistent results, due to solvent evaporation. Use Empower Configuration Manager or MassLynx Plate Generator to define custom plate layouts; the most reliable way to create a custom layout is to modify an existing layout. If you are using Analyst software, you may need to change the default custom plate settings. See the ACQUITY UPLC System Driver release notes ( rA) for more information. Do not use Open Access plate kits (part number or ) with ACQUITY UPLC or H-Class systems. 3 Cap mats and sealing films ACQUITY H-Class Follow these recommendations when selecting cap mats for use with the ACQUITY UPLC Sample Manager and Sample Organizer and the ACQUITY UPLC H-Class Sample Manager-FTN: For optimal reliability when using PEEK needles, do not use resealable cap mats from manufacturers other than Waters. If you do, use 100% stainless steel or stainless-steel-tipped needles. Replace Alliance system cap mats (part number WAT and WAT058943) with ACQUITY UPLC cap mats (part number or , depending on the plate type). Polypropylene cap mats give inconsistent results at 4 ºC. If permissible, raising the sample temperature to between 4 and 10 ºC should improve polypropylene cap mat performance. Cap mats can become brittle when cooled below ambient temperatures. Testing confirms that ACQUITY UPLC cap mats on plates filled with either water or water/methanol (90:10) solution are pierceable and retain closure at temperatures of 4 ºC, 23 ºC, and 40 ºC. Cap mats were tested using a Sample Manager equipped with the standard piercing needle, with a metal tip, or a longer piercing needle, with the standard PEEK sampling needle (updated Y-carriage design and v1.30 or v1.40 ACQUITY UPLC driver). Using incorrect cap mats can result in shredding (with metal tipped needles) or bent needles (with PEEK needles). Shallow plates can deform if the heat sealing process is long in duration. Pre-slit mats perform with greater reliability than solid mats with ACQUITY UPLC and H-Class systems , 30 L PEEK , 30 L FEP with SS tip , 30 L SS with SS tip , 15 L SS with SS tip , 15 L PEEKSil with PEEKSil tip , 30 L PEEKSil with PEEKSil tip , H-Class 15 L , H-Class 30 L Cap mats Polypropylene, for round-well plates, 96 1 ml Polypropylene, for square-well plates, 96 2 ml PTFE/silicone, for square-well plates, 96 2 ml PTFE/silicone, for round-well plates, 96 1 ml 4 Heat seal films and accessories Clear polyester heat seal film, peelable Aluminium foil laminate heat seal film, peelable volt plate sealer volt plate sealer 4 When using cap mats with 96-well (2 ml square, 350 L round, or 1 ml round) plates, set the aspiration rate to the default, automatic, setting (100 L/min.) or slower. 5 Use only stainless steel or stainless steel tip needles with ACQUITY UPLC driver v1.23 or earlier. 4 Vials and septa Follow these recommendations when choosing vials: ACQUITY H-Class Holders Use Waters-certified vials whenever possible. 4 ml vials are not recommended for use with ACQUITY UPLC systems. Do not overfill vials. When using the H-Class Sample Manager-FTN 6, adjust the needle placement setting to no longer than the following values to avoid damaging the needle: Waters Total Recovery vials 0.7 mm Waters Max Recovery vials 2.1 mm Waters flat-bottom vials 0.1 mm To access the setting, click the General tab in the Sample Manager instrument method, and then click Advanced. The Sample Manager s Z-axis calibration affects the needle placement within the vial. Calibrate the Z-axis whenever you change the needle. Pre-slit septa perform better than solid septa with ACQUITY UPLC and H-Class systems. 5 Plate configuration and needle placement For ACQUITY UPLC Sample Managers, use only plate types that are designated ANSI-XXXXXXXXX. Custom plates and holders can be reliably generated by modifying an existing format.
