5 BIOLOGICAL SAFETY CABINET Installation and Operations Manual Revision: 29-Aug-2018 Page 5 of 34 Investigators should consult with a Biosafety officer or qualified safety professional and request a risk assessment of the proposed investigation to ensure that an appropriate BSC is used for the work. Purchase of an NSF 49 Listed Class II biosafety cabinets is recommended, but alternative containment equipment may be suggested for special tacks such as Robotic Enclosures. a. The investigator should thoroughly examine the intended installation site to ensure it will meet the requirements for proper cabinet operation. b. Allow at a gap of at least 6 (15cm) above HEPA to allow for scanning, and to allow at least 6 (15cm) should be maintained on both sides of the BSC, as well as 12 (30cm) behind the unit, to allow for service operations if necessary. c. BSC s should be placed in a position where there should be no opposing wall (or other obstruction likely to affect the airflow) within 6ft (180 cm) of the front access opening if possible. d. BSC s should not be installed in positions where they are not likely to be affected by other items or equipment. In particular the distance to the front access opening of an opposing BSC, fume hood, or the edge of a local exhaust ventilation outlet should not be less than 5ft (150 cm) if possible. e. Any room air supply diffuser should not be within 10ft (300 cm) of the front access opening if possible. f. Doorways should not be within 5ft (150 cm) of the front access opening or within 3ft (90 cm) of the side of the BSC. g. The position of a BSC should satisfy the spatial requirements (e.g. vision, lighting and convenience of access) of the operator and personnel working nearby. When a BSC is installed on a bench top, the leading edge should be flush with or slightly overhanging the edge of the bench top. WORKING ENVIRONMENT: 1. Only suitable for indoor use; 2. Ambient temperature: 15 C~ 35 C; 3. Relative Humidity: 75%; 4. Atmospheric pressure range: 70 kpa~i06 kpa; 5. Electrical parameters: Consistent with the rated voltage of the biosafety cabinet (See Technical Parameters and Performance Index); 6. Power supply needs to have good grounding and preferably operate on its own circuit; 7. Test the voltage stability before using: If the voltage is unstable, the voltage regulator should be used, otherwise the control panel and transformer may be easily damaged. The cabinet must be located in an area protected from air currents that originate from ventilation systems, air conditioners, doors, windows, and personnel movement. Tests have shown that if the drafts or other disruptive currents exceed the intake velocity of the cabinet, then room contamination may enter the sterile cabinet work zone and vice versa. Thus, the proper placement of cabinets in laboratories or work areas is essential. Attention should be paid to the relationship between the cabinet exhaust air discharged from the top of the unit and any room ventilation air inlets or exhaust louvres. Any location which might contribute to the restriction of cabinet exhaust quantity must be avoided. Particular care must be taken with the electrical connections. Each Biological Safety Cabinet must be on its own dedicated circuit and plugged directly into the receptacle. Under no circumstances should an extension cord be used, or the existing electrical cord be modified without consulting the factory. The Biological Safety Cabinet is designed for continuous operation and all electrical circuits and motors are fully protected. If ground fault interrupters are required, we suggest that the duplex outlet into which the unit is plugged be converted to a ground fault interrupter circuit. This is easily carried out at the duplex electrical outlet itself or at the circuit breaker in the electrical control panel. NOTE: Refer to current National Sanitation Foundation Standard No. 49 for recommendations regarding suitable locations for a Biological Safety Cabinet.

11 BIOLOGICAL SAFETY CABINET Installation and Operations Manual Revision: 29-Aug-2018 Page 11 of Installation Before packing and transportation, the Biological Safety Cabinet has been tested in conformance with NSF standards in our factories, but additional on-site tests must be performed. Only qualified Certification Technicians can make any adjustments in order to ensure the accuracy of the on-site testing, test equipment must be calibrated. Please contact the manufacturer to provide names of local certification companies. On-site testing should include at least the following: Leak testing of supply and exhaust HEPA filters using an aerosol generator and calibrated photometer. Check the safety cabinet's inflow and supply velocities. Check airflow inside the cabinet by using smoke simulator; When the cabinet is connected to building exhaust system, balance the exhaust system. Contact your safety committee to provide an on-site inspection as per NSF F.7; the equipment should not be used without this pre-inspection. Do not adjust the fan speed in the absence of fully qualified technician. If the manufacturer has already set the correct air flow, generally it does not require further adjustments Test and standard 1. Filter leakage test: as per NSF F.5 The standard aerosol tests will confirm the integrity of the HEPA filters, generally tested at least annually. Qualified personnel must use calibrated device when aerosol testing. 2. Air adjustment of the opening of the work area: as per NSF F.3 The exhaust HEPA filter is installed on the top of the Biological Safety Cabinet. The air volume exhausted is equal to the air volume entering from the working opening. The average velocity entering the opening of the work area has been listed and outlined as in NSF-49. For the opening air velocity testing data, see APPENDIX 1. To adjust the fan speed, adjustment should be according to the data specified in APPENDIX 1. To set the air velocity of the opening work area correctly. We emphasis again that only qualified technicians using calibrated equipment should be allowed to carry out the adjustments. If there any difficulties, please contact the manufacturer. Note: the adjustments must be made in conjunction with the down flow air velocity as indicated in APPENDIX 1 and it is necessary to have a balance test between them Inflow velocity measuring method: as per NSF F BK3-4 with 8 opening only Adjust the sash opening to normal operating height of 8 and use two rows of points in the plane of the nominal access opening. The top row is 1.97 (50 mm) down from the top of the opening. The bottom row is 1.97 (50 mm) up from the bottom of the opening. Spacing begins 4 (102 mm) from the side walls with 3 (91.44 mm) between points. The armrest remains in place during the test. Measurements are to the center of the white sensing element. The anemometer is positioned vertically, taped to the outside of the sash. The average data is the inflow velocity.

12 BIOLOGICAL SAFETY CABINET Installation and Operations Manual Revision: 29-Aug-2018 Page 12 of 34 BK3-4 with 10 opening only Adjust the sash opening to 3 (76mm) and remove the armrest and tape gap between the work tray and hood frame. Testing point is in the same plane with the window starting 4 (102 mm) from the two side walls. The grid concists of a single row 3.60 (91.44 mm) apart with a total of 12 readings. Set center of probe to centre line 1.5 (38mm) and taped to outside of the sash with probe facing downward for each reading. The correction facture is 1.49 BK3-6 with 10 opening only Adjust the sash opening to 3 (76mm) and remove the armrest and tape gap between the work tray and hood frame. Testing point is in the same plane with the window starting 4" (102mm) from the two side walls. The grid consists of a single row 4.92 (125 mm) apart with a total of 13 readings. Set center of probe to center line 1.5 (38mm) and taped to outside of the sash with probe facing downward for each reading. The correction factor is 1.21 Using Direct inflow measurement: as per NSF F Seal by taping the device to the center of the front opening, seal the open areas on either side of the capture hood portion of the DIM as necessary. Take at least five readings and average them to determine inflow volume rate. Care should be taking not to restrict the airflow through the instrument intake. Calculate the average inflow in cubic feet per minute (cubic metres per second) by the work access opening area to provide the inflow velocity in FPM or (M/S). These calculations have already been documented and are shown in the test sheets provided. 3. Downflow air velocity: as per NSF F.2 The airflows should be balanced and generally no further adjustment is required. How ever when the HEPA filter is blocked the readings on the static pressure monitor increases (located on the control panel). The fan speed needs to be adjusted so that it can maintain the correct air velocity within the working chamber. Balancing can control the exhaust and inflow air ratio by adjusting the exhaust damper position to correct the front to back air split. The setting is done in the factory and generally needs no adjustment. For the correct settings and the test data, please refer to APPENDIX 1. NOTE: Adjusting the balance of the air velocity should be carried out by qualified personnel only. Down flow velocity measuring method: BK3-4 The testing point is from the work area 4 (1OOmm) up from the top of the sash opening in the plane of the normal access opening. The grid consists of 3 rows with 8 points per row, spacing begins mm from the side walls and front glass opening with equally spaced position between points, a square grid front to back 5.12 (130mm) X left to right 5.32 (135inm), the average data from 24 points is the Down flow velocity. BK3-6 The testing point is from the work area 4 (1OOmm) up from the top of the sash opening in the plane of the normal access opening. The grid consists of 3 rows with 11 points per row, spacing begins 6 (150 mm) from the side walls and front glass opening with equally spaced position between points, a square grid front to back 5.60 (142.2mm) X left to right 5.09 (149inm), the average data from 33 points is the Down flow velocity.
