Chapter 3 Stack the Modules RIGOL Chapter 3 Stack the Modules Unpacking the boxes in following order: L3500 UV-Vis detector/l3520 DAD, L3400 column oven, L3320 autosampler, L3210/L3220/L3240/L3225/L3245 high pressure pump, L3100/L3120/L3140 solvent organizer. The number and variety of the module may differ with different system configurations, but should remain the installation sequence. L-3500/L-3520 Detector 1. According to the requirement of Unpacking Procedure, complete the inspection and unpacking. 2. The two transportation screws on the bottom of L3500/L3520 detector depicted in following pictures are intended for transportation purpose, and must be removed before installation. You can find them easily at the bottom plate of detector by the guidance of red arrow beside them. Note: Save those transportation screws properly for future usage. These two screws are larger than T20, remove them with proper driver or hex key. Plate 1 Remove the transportation screws before installation 3. Put the module on the workbench carefully, connect the rubber waste pipe to the drain outlet at the lower right corner of the front panel (under the leak collection tray). Note: It is import to connect the waste pipe now because the waste outlet is hardly accessible after the whole stack was settled. Plate 2 Connect drain pipe L-3000 Installation Reference 3-1

RIGOL Chapter 3 Stack the Modules L-3400 Column Oven 1. According to the requirement of Unpacking Procedure, complete the inspection and unpacking. 2. Put L-3400 column oven on the detector. The supporting leg under the column oven can be put in the dents above the detector, which made the stack relative stable. This works for each module. Note: L3400 does not have transportation screws. L-3320 Autosampler 1. According to the requirement of Unpacking Procedure, complete the inspection and unpacking. 2. Put L-3320 autosampler on the column oven. Note: L-3320 does not have transportation screws. Note: If the system is equipped with manual injection valve (7725i) instead of autosampler, it should be fixed on the right of the pump, you can see three screw holes on the pump reserved for this purpose. In this case, pump should be unpacked before manual valve. Plate 3 Holes reserved for manual valve L-3200 series Pump 1. According to the requirement of Unpacking Procedure, complete the inspection and unpacking. 2. Put L3200 pump on the L3320 auto sampler. Note: There are two transportation screws for L-3210/L-3240/L-3245 while four are there for L-3220/L-3225 (two for each pump body), remove them before stacking with a T20 screw driver. Chapter 4 Tube connection RIGOL Chapter 4 Tube connection Preparation Note: Put on gloves before starting. A. Remove all the adhesive tapes (intended for transportation) on the modules before starting. B. The front door of each module was connected to the housing by magnets on the left and can be removed. In case to do this, open the door from the right, lift slightly and pull to disconnect from the left. Plate 6 Open the front door, lift and pull to disconnect it C. Tubes on the pump was already fixed, other tubes between different modules, solvent bottle and the waste require user s work. These tubes are pre-cut, shaped and can be used directly. Find these tubes in the accessory boxes for each module. Note: Do not over-tighten the nuts. For stainless steel screws, tighten it first by hand. The screw should go into the connector smoothly, some resistance can be caused by improper angle between screw and connector, remove it and try again in this case. When the screw reach the end and cannot be turned by finger anymore, tighten it no more than 90 degrees with a wrench. Usually 90 degrees is enough, sometimes a little more is required to stop leaking. But NEVER THIGHTEN THE STAINLESS STEEL NUT MORE THAN 180 DEGREES WITH WRENCH. Binary high-pressure gradient pump: 1. Remove the front door of solvent organizer and pump. 2. Make the transparent FEP tubes through the bottle caps and connect them to the frits (metal filter), connect the other side with PEEK nuts to online degasser in L3100 solvent organizer. 3. Use the shorter FEP tubes (which have PEEK nuts on both end) to connect online degasser with inlet check valves under the right pump head. Note: Use transparent FEP tubes to connect solvent bottles and inlet check valves under the right pump head directly if the system not configured with online degasser. Note: The blank nuts used for sealing the system should be kept properly for future usage. Note: Pay attention to channel tag on the tube and the label on pump. For binary system, the pump on the right side was designated as pump A and should be connected to tubes with tag A on them. 4-4 L-3000 Installation Reference

Chapter 4 Tube connection RIGOL B. Quaternary low-pressure gradient pump 1. Remove the front door of solvent organizer and pump. 2. Make the transparent FEP tubes through the bottle caps and connect them to the frits (metal filter), connect the other side with PEEK nuts to online degasser in L-3140 solvent organizer. Note: For system with L-3245 quaternary pump, online degasser is integrated in the pump module, in this case L-3140 is not necessarily exit. 3. Use the shorter FEP tubes (which have PEEK nuts on both end) to connect online degasser and four channel proportion valve. Note: These tubes were pre-install for L-3245 quaternary pump, not need for user s work. Note: Pay attention to channel tag on the tube and the label on pump. For quaternary pump, channel label can be found on four channel proportion valve. C. Seal wash tube and purge drain tube 1. Take out three transparent FEP tubes which are different in length, use them properly. 2. Use these three tubes to connect washing bottle to peristaltic pump, pump, A pump and washing bottle respectively. 3. Connect the FEP tube to the outlet port of purge valve (below the valve), the other end should be plug into a suitable reservoir for waste. Note: RIGOL recommend aqueous solution with 10% isopropanol as seal wash solvent, due to its good solubility to salt. The solution should be freshly made and replaced every day. Tube connection of autosampler 1. Remove the front door of autosampler. 2. Connect purge valve outlet (on the right of the purge valve, where an online filter can be found) to Port 3 on six ports valve of autosampler with a pre-shaped stainless steel tube. Note: If manual injection valve was used instead of autosampler, connect purge valve outlet to Port 2 on manual injection valve. 3. Connect Port 2 on six port valve to the connector on column oven. Note: Port 3 should be connected to connector on column oven in case of manual injection valve was used. 4. Put out washing tube in L3320 autosampler accessory kit, connect the PEEK nut end to washing port on four ways valve in autosampler, and connet the other end with the frit, put it in adequate washing buffer (Normally use methanol for the first time due its ability to remove bubbles. Solvent with good solubility to sample should be used in future usage. 50% methanol aqueous solution is a good choice if you are not sure about the solubility). 5. Find the waste tube in L3320 autosampler accessory kit (it look exactly like the washing tube), connect the PEEK end to the right port on four ways valve, let the other end go into the waste tank. 6. Connect the bold rubber pipe to the right waste port on the autosampler, put the other end into waste. L-3000 Installation Reference 4-5

RIGOL Chapter 4 Tube connection Tube connections from column oven to flow cell in detector 1. Install column. Note: Check the flow direction, mobile phase go through the column oven from left to right, columns should be installed in accordance with this flow direction. Note: The highest sustainable pressure of PEEK nut on the left side of chromatographic column is 2000 psi, you should use stainless steel fitting(in the accessory kit) instead of PEEK one when the system pressure is higher than the limit. Note: The system cannot work steadily except that the front door of column oven is closed tightly. Pay more attention to the groove on the right side of the front door, the tube inlet and outlet of the column oven must be compatible with it, otherwise the alarm will be triggered once the door not closed tightly. 2. Connect column outlet and flow cell inlet in detector with the pre-cut PEEK tube in accessory kit. Note: Elutes go into flow cell from the lower inlet, the higher one is outlet. Tube connections from flow cell outlet to the waste Connect the white outlet tube to the outlet port of flow cell (upper one), the tube is about 80cm long (0.17mm inner diameter) to provide essential backpressure for the cell. 4-6 L-3000 Installation Reference

Put all the power switches off, tidy up the power cable at the rear position of the modules, and keep them clean. L-3000 Installation Reference 6-1

错误! 未找到引用源 错误! 未找到引用源 RIGOL Chapter 7 Clarity installation Note: Install RIGOL CWS before connecting any device and dongle. GET YOUR COMPUTER READY Before installing workstation, make sure your computer comply with the Pre-Installation Guide, and has the following software installed: Microsoft windows XP SP3 or above, or Windows7. Microsoft.Net Framework 3.5 or above. NI-VISA 4.6.2 or above. You can download NI-VISA here: http://joule.ni.com/nidu/cds/view/p/id/1605/lang/en/ Note: NI-VISA must be installed before RIGOL CWS/Clarity, or the L3320 autosampler driver will be unavailable. INSTALL WORKSTATION Install Clarity with the CD enclosed in the package. 1. Install Clarity before connecting any device. 2. Connect dongle key to USB port, the driver should be installed automatically. 3. Start Clarity, complete configuration process. 4. Log into Clarity and check the connection, if everything goes well, you can use RIGOL HPLC system now. L-3000 Installation Reference 7-1

Chapter 8 Rinse the system RIGOL Chapter 8 Rinse the system Get the solvent ready, connect a syringe at the end of purge waste tube, open purge valve a little bit and draw to clean all the bubbles in each channel. Note: For binary pump, the solvent in the bottle should go slowly into the system and rinse the air out for the two channels simultaneously. If this doesn t happen, there could be something wrong with the check valve, an ultra-sonic bath for the check valve will solve this problem. Note: If a quaternary pump is installed, solvent will not go through the tube because the proportion valve is closed by default. In this case the system must be wired up and communicates with workstation, set a flow for each channel (so the pump will open the corresponding channel in proportional valve) and then draw, clean all four channels one by one. The following steps can only be done with the help of workstation: 1. Rinse all channels one by one with methanol at flow rate 3mL/min. 2. Do initial washing for autosampler. Get ready for use Now you can purge the system, equilibrate the column and start you chromatography experience with RIGOL L-3000 HPLC. L-3000 Installation Reference 8-1
