absorption trap Operation: Absorption of noxious acid fumes and gasses without mechanical fume extraction. COLD SOLUTIONS When substantial quantities of a solid are to be filtered from suspension in a liquid, a Buchner funnel of convenient size is employed. Suction causes rapid filtration and results in a more complete removal of the base liquid than in normal atmospheric filtration. Select a filter paper of such size that it covers the entire perforated plate without being folded up against the sides of the funnel (it should be slightly smaller than the plate). Moisten the filter paper with the liquid to be filtered, place it on the perforated plate and apply the suction. The filter paper will adhere firmly to, and completely cover, the plate of the funnel thereby preventing any solid matter from passing underneath the filter paper and into the flask. Pour as much of the liquid into the funnel and allow it to be filtered before transferring the residual slurry into the funnel for filtration. This method allows quicker filtration than if the mixture had been stirred to bring more particles into suspension and pouring it directly unto the funnel. Initial gentle suction leads to more effective filtration than powerful suction which could cause finer particles of solid to be drawn into the pores of the filter paper reducing the rate of filtration. After completion of a reaction, filtration of a mixture will often be necessary to either isolate a solid which has separated out or remove insoluble impurities or reactants, in which case the desired material remains in solution. Any solid remaining in the reaction flask can be easily transferred to the filter funnel by rinsing with a bit of the filtrate and stirring well to remove any solids adhering to the sides of the reaction flask. Be sure to disconnect the pump from the Buchner flask before switching off the pump otherwise suck back will occur and ruin any reaction products in the filtrate. In either case, the lid of the desiccator should be slightly ajar for 30 seconds prior to complete closing. This will prevent a partial vacuum from forming as the heated air cools. If such a vacuum forms it might become very difficult to remove the lid without upsetting the samples within. A dried sample should remain in the desiccator for at least 5 minutes before being weighed. If the sample is to remain in the desiccator for an extended period of time before weighing, the top of the weighing bottle of the lid of the vial should be put in place after 10 minutes of cooling. An airtight seal is maintained by applying a small amount of silicone grease to the flanged surfaces where the lid and body of the desiccator meet. Magnetic stirring and mixing is used in a number of common laboratory procedures. The importance of selecting the best stirring bar for each particular application is often overlooked. Varying the size, shape or length of the magnetic stirring bar will after the motion imparted to the solution. Thus, selection of the appropriate stirring bar for given application should be done with great care. Understanding the principles of magnetism is important. On the most basic level, we are aware that opposite poles attract. However, we must also understand the significance of the drive element, magnetic coupling, spin-out, vessel shape, viscosity and turbulence. The drive motor of a magnetic stirrer provides the rotation necessary to spin the drive magnet. The drive magnet is generally a bar or U-shaped alloy or a ceramic disk. The magnetic poles are typically about 50mm apart in bench top models. The poles can be up to 140mm apart in units large enough to mix 190litres of liquid solution. For optimum efficiency and magnetic coupling, the distance between the magnetic poles of the drive magnetic poles of the drive magnet and the length of the stirring bar should be equal. Therefore, the drive magnet dictates the longest possible stirring bar that can be used effectively. A small metal washer moved around the surface plate of the stirrer will naturally gravitate towards one of the poles of the drive magnet. A small magnetic stirring bar will provide similar results. Another important parameter in choosing a magnetic stirring bar is the vertical distance between the drive magnet and stirring bar. The material and thickness of the cover plate as well as that of the containing vessel can also influence stirring efficiency. Cover plates are made of paramagnetic materials such as ceramic or aluminum, which offer better coupling of the drive magnet and stirring bar. The magnetic energy is not impeded as it might be if the top were made of magnetic materials. The amount of magnetic energy that joins the two magnets (coupling) falls off as the distance between them increase. Stated mathematically, the coupling energy of force is inversely proportional to the square of the distance between them. It stands to reason that for the best magnetic coupling, the distance between the magnets should be minimized. Magnetic stirring bars are generally made of ALNICO (an alloy of aluminum, nickel, iron and cobalt) encapsulated in an FDA approved PTFE coating. This combination offers the best compromise of magnetic strength, usefulness and cost. The PTFE coating is only useful at temperatures up to 225 C. Other coatings or coverings such as glass tubing, polypropylene and polyethylene are also used. More exotic materials have not been found to be commercially viable. High energy materials, such as Samarium Cobalt and Neodymium Iron Boron, though having a much higher energy product (Bd x Hd) than ALNICO V, do not always work better as drive magnets. The geometry of the magnet, in the case of bar magnets, length/diameter (L/D), governs how effective the magnet will be. The aforementioned magnets have energy products of three to six tomes that of ALNICO V in geometry of 1 to 1. When used in a 4 to 1 configuration, their relative levels are nearly the same or less than the ALNICO V. Additionally, many of the exotic materials are extremely temperature sensitive and will demagnetize as the temperature increases. Once a magnetic stirring bar has been placed in a container with a solution, it should be positioned directly over the centre of the drive magnet. The stirring speed should be increased slowly, until the desired vortex pattern is achieved. Should the magnetic stirring bar lose its coupling with the drive magnet because of the speed of the drive magnet or the viscosity of the fluid, it is said t have spun-out. Improperly selected stirring bars are the frequent cause of spin-out, as well as a drive magnet speed that is too fast. Matching the drive and stirring magnet improves results dramatically. The selection of the shape of the magnetic stirring bar also influences the resulting vortex. 626

A new range of NMR tubes has now been developed which caters for all the quality criteria for obtaining optimum spectra that were only conceivable before. What is so interesting about NMR tubes that it is worth writing about? Now all NMR users know how very much the quality of their spectra depend on the quality of the sample vessel they are using. There are initially two reasons for this: The sample tube is also measured as part of the measuring process. This means that its material and geometry properties should have preferably no influence on the measurement or at least an influence which can be eliminated by calculation. The sample vessel itself is in an inaccessible position in the measuring instrument during the measurement process, namely in the middle of the magnet. It should not be disturbed there under any circumstances since it is very difficult to interfere with it in this position. The material and dimensions of NMR tubes are subject, therefore, to very stringent requirements and they are accordingly expensive to manufacture. Before a new range of NMR sample tubes comes on the market, a large number of tests have to be carried out before the products agree with the specifications laid down. Then, when they are in full production, measures have to be implemented to ensure that the specifications laid down are adhered to. NMR quality criteria The decisive factor for the quality of a spectrum is how accurately the magnetic field is adjusted to the sample in the NMR tube. As this adjustment (or shimming) can be a complicated process, both the absolute geometry of the tube and the comparability of different tubes are a important quality criteria. For this reason, the adjustments between the different tubes, e.g. in the sample changer, should be as identical as possible. Shimming is then only required once and not for each tube. From this the various quality criteria for a good NMR tube can be derived: Identical sample quantities should come to the same level in several tubes in the sample changer. Otherwise the magnetic field must be readjusted. All tubes should have the same diameter as far as possible. The wall thickness should be the same at every point. The tube must be straight. All the tubes must sit firmly in the spinner without moving and must be a good fit. The tubes should be robust so that they cannot be damaged in the NMR unit. The glass should be chemically resistant and inert to all samples. The glass may not contain any paramagnetic impurities that can result in problems in the spectrum. The tubes must be labeled so that different qualities can be clearly distinguished. Nowadays, many samples are measured without rotating them as in former times. The reason here too can be found in the homogeneity of the magnetic field. If they are not rotated completely evenly the result is a worse spectrum. It is, therefore, better in many cases to compensate for any irregularities by shimming. However, it is even better to ensure that as few irregularities as possible occur with an accurately specified quality. Most NMR spectroscopy workers have got used to rolling NMR tubes on a flat surface as a rough test. This way of sorting out the bananas makes sense as a preliminary test in practice, but of course it is not adequate for production development and production control purposes. 637
