3 1. Attach the Bottom-of-the-Bottle pump inlet filter (# ) to a length of 1/8" OD PTFE tubing (# ) using the flangeless nut and ferrule supplied with the inlet filter. Refer to fig. 2 for instructions on making flangeless fitting connections. 2. Insert the pump inlet filter with attached tubing into the reservoir until the inlet filter touches the bottom of the reservoir. Mark and cut the tubing 1/2" above the point where the tubing exits the top of the reservoir. 3. Connect the tubing to the Pump Port (see fig. 3) on the bottom of the cap using an ETFE flangeless nut (# ) and a yellow ETFE ferrule (# ). 4. Attach the sparge inlet filter (# ) to a second length of 1/8" OD PTFE tubing using the Plastictight nut supplied with the inlet filter. 5. Insert the sparge inlet filter with attached tubing into the reservoir until the bottom of the inlet filter is above the position of the pump inlet filter. This prevents the helium sparge bubbles from being drawn into the pump. Mark and cut the tubing 1/2" above the point where the tubing exits the top of the reservoir. 6. Connect the tubing to the Sparge Port (see fig. 3) on the bottom of the cap using an ETFE flangeless nut (# ) and a yellow ETFE ferrule (# ). Figure 2. Flangeless Fittings Instructions Ferrule Nut Tubing 1. Cut the tubing, leaving a square-cut face 2. Slide the nut onto the tubing. 3. Slip the ferrule onto the tubing with the conical section toward the nut. The flat end of the ferrule should be even with the end of the tubing. 4. Insert the tubing with ferrule into the mating fitting, making sure the tubing is firmly pressed against the flat surface of the fitting. 5. Tighten the nut. 1. Attach the Bottom-of-the-Bottle pump inlet filter (# ) to a length of 1/8" OD PTFE tubing (# ) by slipping the stem of the inlet filter as far as possible into the ID of the tubing. 2. Insert the inlet filter with attached tubing into the reservoir until the inlet filter touches the bottom of the reservoir. Mark and cut the tubing 1/2" above the point where the tubing exits the top most portion of the reservoir. 3. Connect the tubing to the Pump Port (see fig. 3) on the bottom of the cap using a stainless steel flangeless nut (# ) and a yellow ETFE ferrule ( ).

4 4. Attach the sparge inlet filter (# ) to a second length of 1/8" OD PTFE tubing (# ) by slipping the stem of the inlet filter as far as possible into the ID of the tubing. 5. Insert the sparge inlet filter with attached tubing into the reservoir until the bottom of the inlet filter is above the position of the pump inlet filter. This prevents the helium sparge bubbles from being drawn into the pump. Mark and cut the tubing 1/2" above the point where the tubing exits the top of the reservoir. 6. Connect the tubing to the Sparge Port (see fig. 3) on the bottom of the cap using a stainless steel flangeless nut (# ) and a yellow ETFE ferrule (# ). Lower the two inlet filters into the HPLC reservoir and screw the cap onto the reservoir by tightening the blue lower screw clamp. 8. Place the glass support frit into its cavity on the top of the cap. 9. Center a 47 mm membrane filter on the top surface of the cap. The membrane filter and support frit must be dry. 10. Carefully slide the solvent pickup adapter into place on the top of the cap and clamp together by tightening the blue upper screw clamp. Be careful not to disturb the filter since it acts as the vacuum gasket during filtration. 11. Place the blue BEVEL-SEAL open top cap followed by the o-ring over one end of the 1/4" OD PTFE tubing. 12. Insert the tubing as far as possible into the BEVEL-SEAL thread on the side arm of the solvent pickup adapter and tighten the cap until snug. 13. Cut the 1/4" OD tubing so that it will just reach the bottom of the vessel containing the unfiltered mobile phase solvent.

5 14. Screw the 1/4-28 to male luer adapter (# ) into the vent/vacuum port on the back of the cap (see fig. 4). Connect the 1/4-28 to 1/4" hose connection adapter (# ) to the 1/4-28 to female luer adapter (# ). 16. Insert the 1/4" hose connection adapter into the vacuum source tubing. The vacuum source tubing must have a 1/4" ID. If a vacuum pump is used, an appropriate vacuum trap inline between the cap and the vacuum pump is recommended. Please refer to for our complete listing of vacuum traps. 17. Slip the female luer with vacuum tubing onto the male luer at the vent/vacuum port. 18. Close the HI-VAC valve on the top of the solvent pickup adapter. Close the PUMP, SPARGE and RECIRCULATE (Five Valve Caps only) valves on the cap by rotating clockwise. Open the VENT and FILTER valves by rotating counter-clockwise. 19. Start the vacuum pump. 20. When a vacuum has been established in the reservoir, open the HI-VAC valve on the top of the solvent pickup adapter. The unfiltered mobile phase solvent should rush through the tubing into the solvent pickup adapter and cover the filter membrane to a depth of several millimeters which will remain at approximately that depth until filtration is complete. If the filtration needs to be temporarily interrupted, close the FILTER valve. This maintains mobile phase solvent on top of the membrane filter and allows the filtration to be easily restarted. 21. When the filtration is complete and all of the mobile phase solvent has been drawn into the reservoir, close the HI-VAC valve on top of the solvent pickup adapter to vacuum degas the mobile phase. After filtration and vacuum degassing are completed, close the FILTER valve. 22. Connect the SPARGE port on the back of the cap to the helium source using the 1/8" OD PTFE tubing (# ), an ETFE flangeless nut (# ) and a yellow ETFE ferrule (# ). 23. Open the SPARGE valve to allow helium to enter the reservoir. Caution: Do not allow the pressure in the reservoir to exceed 6 psi. Open the VENT valve to allow the venting of the sparged gasses. Directing these exhaust gasses to an appropriate vent hood can be accomplished by connecting a length of 1/8" OD PTFE tubing to a 1/4-28 to female luer adapter (# ) with an ETFE flangeless nut (# ) and a yellow ETFE ferrule (# ). The female luer is then slipped onto the male luer adapter in the VENT/VACUUM port. 24. When sparging has been completed, close the VENT valve to blanket the mobile phase solvent and prevent the reintroduction of unwanted gasses into the mobile phase.

6 25. Connect the PUMP port on the back of the cap to the LC pump (see fig. 5). 26. Open the PUMP valve to allow the mobile phase to be drawn from the reservoir by the pump. Between HPLC runs the mobile phase can be isolated in the reservoir by closing all of the valves on the cap. 27. (Five Valve Caps only) Connect the RECIRCULATION port on the back of the cap to the HPLC detector or automatic solvent recycler using flangeless fittings with 1/4-28 threads. Remove the blue upper screw clamp and glass support frit from the cap. 2. Screw the 90 mm Conversion Base (# ) onto the cap. 3. Place the supplied 90 mm glass support frit into its recess on the top of the conversion base. 4. Center a 90 mm diameter membrane filter on the top surface of the conversion base. The membrane filter and support frit must be dry. 5. Carefully slide a 90 mm solvent pickup adapter (# or # ) into place on the top of the conversion base and clamp together by tightening the black upper screw clamp. Be careful not to disturb the filter since it acts as the vacuum gasket during filtration.
