This introduction may be greatly reduced by adding several key components to the typical system. These are: a properly sealed solvent reservoir, a solvent waste container with secure fluid connections, and a carbon filter to capture vapors from both reservoir and waste when a fume hood is unavailable. These components reduce health hazards and simplify solvent disposal while satisfying the requirements of government regulatory agencies. 3 1 Solvent reservoirs and caps Standard laboratory bottles and closures The typical reservoir used for laboratory solvents at the inlet of an HPLC is either a 1 liter glass bottle, with a 45mm cap size (commonly referred to as GL45), or the standard 4 liter amber glass bottle in which the solvents were shipped (which measures 38mm across the threads, and referred to as GL38). 7 2 Waste container and manifold Secure connections reduce laboratory hazards The industry standard for waste containment in the laboratory is rapidly changing. Fire-marshal approved methods which minimize risks to health and safety from fire, chemical burns, or toxin inhalation are necessary measures made possible only by proper fluid connections. 8 2a Waste containers 4 LITER BOTTLES AS SOLVENT WASTE CONTAINERS Standard 4liter bottles are often considered an acceptable means of waste containment. A sealed bottle cap or bottle cap manifold may be used to properly connect waste lines (see Section 1a: GL38 bottle caps). Use a polyethylene sleeve, part number JT-12009, to contain the glass bottle in case of accidental drops or spills. JT-PP gallon polyethylene safety can with two polypropylene quick disconnects. Fire-marshal approved. Translucent polyethylene safety containers have a built-in pressure relief to release pressure in extreme temperatures. Quick disconnects offer simple detachment of the can from the HPLC without leaks, drips or vapor release. Once disconnected, the can is automatically sealed to prevent hazardous spills. 9 2b Waste manifolds INLET / WASTE MANIFOLDS The manifolds below, part numbers BC and BC , are for use with bottles only. All wetted surfaces are PTFE, including the tubing adapters, which makes these manifolds ideally suited for the harshest of laboratory chemicals. The fifth port is occupied by a barbed fitting for use as a vent. Attach 1/4 ID tubing to this barb and connect to a carbon vent filter (section 3a) or fume hood. 10 2c Waste manifold compression fittings A NOTE ON COMPRESSION FITTINGS AD Compression fitting for micron. Usually more secure than barbed fittings, compression fittings grip hard or softwall tubing with a ferrule. They will keep tubing firmly connected to waste manifolds. Use compression fittings especially on hard-wall tubing or tubing of smaller diameter than 1/8 or 1/16. To secure, slide first the fitting and then the ferrule over the tubing. Insert the assembly into the NPT adapter. Tighten the nut firmly. The more tightly the nut is threaded, the better the ferrule will grip the tubing. AD Compression fitting for 1/8. 12 3 Toxic solvent vapor filter Activated carbon filter for organic vapors Fume hood space in the laboratory is often limited, and the plumbing required to pipe an emissions source to the hood can be cumbersome to install. With a small benchtop carbon filter, toxic vapors from multiple sources may be dealt with at or near the emission source. The activated carbon is supplemented by a color indicator, which changes color from pink to black when the cartridge is spent. The filter assembly includes one carbon filter with a benchtop stand, and either a four- or eight-port manifold.
