SECTION 1 Contains system specifications and a brief physical and functional description of the centrifuge, including the operating controls and indicators DESCRIPTION Centrifugation Principle Centrifugation is a process for the separation of heterogeneous mixtures of substances (suspensions, emulsions or gas mixtures) in to their components. The mixture of substances, which rotates on the circular path is subject to centripetal acceleration that is several times greater than the gravitational acceleration. Centrifuges use the mass inertia inside the rotor chamber for separating the substances. Due to their higher inertia, particles or media with a higher density travel outwards, in doing so they displace the components with a lower density, which in turn travel towards the centre. The centripetal acceleration of an object inside a centrifuge as the effect of centripetal force, depends on the distance between the object and the axis of rotation as well as on the angular velocity. It increases linearly as a function of the distance with regard to the axis of rotation and quadratic ally as a function of the angular velocity. The bigger the radius in the rotor chamber is and the higher the speed is, the higher the centripetal acceleration is. However the forces, acting on the rotor also increase. 9

Specifications Rota 4R-V/Fm is a new generation high speed microprocessor controlled centrifuge. It employs a silent brush-less, asynchronous motor freeing the user from periodic brush change and carbon dust. The centrifuge assembly has a solid steel casing. The centrifuge lid is made of heavy sheet steel and cased with thermal insulation. Special hinges used provide counter balancing of the lid in all positions and allow the user to load or unload the rotor without holding the lid. The centrifuge has a steel armoured chamber. 12 1.2 SAFETY DEVICES Lid Lock Device The centrifuge can only be started when the lid is properly closed. The lid can only be opened when the rotor has stopped. If the lid is opened by way of emergency release system during operation, the centrifuge will immediately come to rest while rotor is at run system. If the lid is open, the drive is completely separated from the run system & another function Standby Mode Monitoring System Entering to setup mode is only possible if the rotor is at a standby. This standstill is checked by the microprocessor Imbalance Monitoring System The indication imbalance centrifuge is in the impermissible imbalance range. The drive will be switched off in the acceleration phase or during the run Motor overheat Monitoring System Microprocessor continually check motor temperature, if motor temperature rises above permissible level centrifuge automatically deaccelerate. 1 1

1.3. CENTRIFUGE CHASSIS Housing The centrifuge housing is made of MS over a MS chassis and is finished with urethane paint. The control panel overlay is made of MS. Poly Urethane Form insulation outer wall of S.S. chamber covered with M.S. Cabinet as a cooling chamber Door The structural MS door has solenoid lock system, prevents run initiation unless the door is closed and latched. When a run is in progress, In the event of a power failure, the door lock can be manually tripped for sample recovery (TROUBLESHOOTING) Rotor Chamber The S.S. solid chamber covered with M.S. cabinet rotor chamber. The silicon rubber seal cap surrounding the drive shaft are visible in the chamber bottom. A around the chamber top assures sealing by rubber gasket Drive The asynchronous single-phase direct-drive motor is brushless for clean, quiet operation. A tie-down S.S. Allen bolt is used to attach the rotor with S.S. Shaft Adaptor to the drive shaft. The resilient suspension ensures that loads will not be disturbed by vibration, and prevents damage to the drive shaft if an imbalance occurs during centrifugation. Maximum braking may be selected to reduce deceleration time, allowing fast processing of samples; alternately, delicate gradients may be preserved using slower de-acceleration. 1 2

Balance the samples properly refer the Instruction Manual. Make sure nothing is fallen in the chamber The centrifuge makes noise and vibrates slightly at a particular speed while the motor is accelerating and or decelerating Critical speed Vibrations This is normal, all centrifuges have critical speeds at which Slight vibrations take place. avoid running the centrifuge at critical speed either run it at slightly higher or lower speed 3 7

4.2. DO S AND DON TS CENTRIFUGE USERS Do s Always run with a full set of buckets. The rotor should be evenly loaded by distributing the load Balance opposite buckets to an accuracy of within 1 gram while using a swing out rotor and within 0.1 grams while using an angle rotor Don ts Never run a swing out rotor partially loaded If there is a spillage, clean and decontaminate your rotor, chamber, buckets and adapters, regular cleaning will increase the life of your centrifuge. Use a brush with soft bristles for cleaning rotor cavities, tubes and shields Do not use a brush with metal wire bristles which might damage the anodized surface of the rotor cavities. To prevent corrosion after cleaning, dry out buckets and rotors by keeping them upside down on a soft absorbent cloth or a soft surface to drain out excess water. Before every run ensure that the rotor is secured properly to the motor shaft by tightening the Allen screw in clockwise direction by the Allen driver provided with the centrifuge Always remove any broken glass, broken tubes or debris from the centrifuge chamber before operating the centrifuge to avoid damage to the rotor and the centrifuge motor shaft For refrigerated centrifuges, after its use at low temperatures, the centrifuge lid should be left open to allow moisture to evaporate to prevent corrosion of the bowl and the motor bearings. Rotors or buckets should be replaced if any cracks or deformation is observed after years of use Never force open the centrifuge lid while the rotor is spinning Also do not ever try to stop a spinning rotor by hand which could be potentially dangerous and can cause grievous injury to the operator. 3 8

4.3. ACCESSING THE ROTOR IN CASE OF POWER FAILURE In the event of a power failure, it may be necessary to trip the door-locking mechanism manually to remove the rotor and retrieve your sample. Action Turn the power switch to OFF and disconnect the power cord from the main power source. Wait for 20 to 25 Minutes. Result No displays or LEDs are lit. No sound or vibration comes from the centrifuge. Make sure that the rotor is not spinning WARNING If the rotor is still spinning, close the LID and wait until it stops before attempting to remove it. Never try to slow or stop the rotor by hand. Insert the supplied lid remover tool into the override key slot in the front side panel. 3 9

Section 5 Contains procedures for routine operator care, maintenance & warranty, as well as a brief list of supplies and replacement parts Care and Maintenance For maintenance not covered in this manual, contact Plasto Crafts Field Service. WARNING Any maintenance procedure or servicing of this equipment that requires removal of any covers can expose parts that involve the risk of electric shock or personal injury. Make sure that the power switch is OFF and the centrifuge is disconnected from the main power source, and refer such servicing to qualified personnel GENERAL MAINTENANCE Perform the following procedures regularly to ensure satisfactory performance and long service life of the centrifuge. At least once a week (depending on usage) inspect the interior of the rotor chamber for accumulations of sample, dust, and glass particles from broken sample tubes. Clean as required (see CLEANING, below), as these accumulations can result in rotor vibrations. To prevent the rotor from sticking always remove rotor from chamber & clean it Clean the chamber after work is over 5.2. MAINTENANCE OF CENTRIFUGE Unplug the mains power plug before cleaning Carefully remove all liquids, including water and particularly all the solvents, acids and alkaline solutions from the chamber using a cloth in order to avoid damage to the motor bearings. If the centrifuge has been contaminated with toxic, radioactive, or pathogenic substances, clean the rotor chamber immediately with a suitable decontamination agent. ( Please refer to decontamination) Use soap water or other water-soluble, mild cleaning agents with a ph value between 6 and 8 for cleaning the centrifuge and accessories Avoid corrosive and aggressive substances Do not use solvents 4 0

Do not use agents with abrasive particles Do not expose the centrifuge and rotors to intensive UV radiation or thermal stress (e.g. by heat generation) Check the condenser at least once a month for dirt and clean it if necessary. If you have any queries, please contact Plasto Crafts service (see chapter service contact ) Maintenance of Rotors and Adapters Rotors, buckets and carriers are produced with the highest precision, in order to withstand the permanent high stress from gravitational fields. Chemical reactions as well as stress-corrosion (combination of oscillating pressure and chemical reaction) can affect or destroy the metals. Barely detectable cracks on the surface can expand and weaken the material without any visible signs. Check the material regularly (at least once a month) for Cracks Visible damage Pressure marks Signs of corrosion Other changes Check the bores of the rotors and multiple carriers. Replace any damaged components immediately for your own safety Maintenance of Centrifuge Chamber DANGER In order to completely remove the glass particles and metal dust from the rotor chamber Grease the upper third of the rotor chamber with e.g. Vaseline Then, let the rotor rotate for a few minutes at a moderate speed (approx.2000 rpm). Metal particles will now collect at the greased part. Remove the grease and metal particles with a cloth. If necessary, repeat this procedure Sterilisation and disinfection of the rotor chamber and accessories If dangerous materials (e.g. infectious and pathogenic substances) are used, the centrifuge and accessories must be disinfected. 4 1

Load bearing bolts Only greased load bearing bolts ensure the even swinging of the buckets and, therefore, the quiet run of the centrifuge. Non-greased bolts can lead to a system shut-down due to imbalances. Apply a small quantity of grease to the load bearing bolts of the rotor and buckets after each cleaning Sterilisation and disinfection of the rotor chamber and accessories Use commercially-available disinfectants such as, for example, or drugstores). The centrifuge and the accessories consist of various materials. A possible incompatibility must be considered. Before using cleaning or decontamination agents that were not recommended by us, contact the manufacturer to ensure that such a procedure will not damage the centrifuge. For autoclaving, consider the continuous heat resistance of the individual materials (see chapter "Autoclaving"). Please contact us if you have any queries (see "Service contact") Autoclaving The service life of the accessories essentially depends on the frequency of the autoclaving and use. Replace the accessories immediately when the parts show changes in colour or structure or in the occurrence of leaks etc. During autoclaving, the caps of the tubes must not be screwed on in order to avoid the deformation of tubes. 4 2

NOTE Before using any cleaning or decontamination methods except those recommended by the manufacturer, users should check with the manufacturer that the proposed method will not damage the equipment SERVICE The centrifuge, rotor and accessories are subject to high mechanical stress. Thorough maintenance performed by the user extends the service life and prevents premature failure. This is why we recommend having the centrifuge checked by the manufacturer once per year in the operating state and once every three years in the dismantled state. Rubber parts should be replaced after three years. Use soap water or other water-soluble, mild cleaning agents with a ph value between 6 and 8 for cleaning the centrifuge and accessories Avoid corrosive and aggressive substances Do not use solvents Do not use agents with abrasive particles Do not expose the centrifuge and rotors to intensive UV radiation or thermal stress (e.g. by heat generation) DANGER 5.4. CLEANING In the event of service work that requires the removal of the panels, there is a risk of electric shock or mechanical injury. Only qualified specialist personnel are authorised to perform this service work Frequent cleaning will ensure proper operation and prolong the life of the centrifuge. Always clean up spills when they occur to prevent corrosives or contaminants from drying on component surfaces. Remove the rotor from the centrifuge weekly and clean the bowl, drive shaft, shaft cavity, threads, and the tie-down nut using a mild detergent and a soft brush. Dilute the detergent 10 to 1 with water. Rinse thoroughly and dry completely. Lubricate the drive shaft with silica gel after cleaning. 4 3

Keep the interior of the rotor chamber clean and dry by frequent wiping with a cloth or paper towel. To prevent accumulations of sample, dust, and/or glass particles from broken sample tubes from damaging rotor pins (causing rotor vibrations), remove the rotor and clean the centrifuge chamber daily. Spray the centrifuge chamber with an antistatic solution and wipe it clean. (Or use an antistatic wipe.) Clean the centrifuge case and door by wiping with a damp cloth or washing with detergent solution. Do not use acetone or other solvents. Remove the air-intake filters and inspect for dust and dirt. Wash them using a mild soap or detergent. Rinse thoroughly and squeeze excess water out, then air-dry before reinserting Glass Breakage In the case of glass breakage, immediately remove all glass particles (e.g. with a vacuum cleaner). Replace the rubber cushions since even thorough cleaning will not remove all glass particles. Glass particles will damage the surface coating (e.g anodising) of the buckets, which will then lead to corrosion. Glass particles in the rubber cushions of the buckets will cause glass breakage again. Glass particles on the pivot bearing of the load-bearing bolts prevent the buckets and carriers from swinging evenly, which will cause an imbalance. Glass particles in the rotor chamber will cause material abrasion due to the strong air circulation. This metal dust will not only pollute the rotor chamber, rotor, and materials to be centrifuged but also damage the surface of the accessories, rotors and rotor chamber Decontamination If the centrifuge and/or accessories are contaminated with radioactive or pathogenic solutions, perform appropriate decontamination procedures be sure the decontamination method will not damage any part of the centrifuge. Take suitable precautions for your own safety if there is a risk of toxic, radioactive or pathogenic contamination. 4 4

TUBE BREAKAGE If a glass tube breaks, and all the glass is not contained in the bucket or rotor, thoroughly clean the interior of the chamber bowl. CAUTION Be careful when examining or cleaning the gasket or chamber, as sharp glass fragments may be embedded in their surfaces. Examine the gasket to make sure that it is free of glass particles. Carefully remove any glass particles that may remain. If glass is under the gasket, call Plasto Crafts Field Service to replace the gasket. Carefully wipe away any glass particles that remain in the bowl STORAGE AND TRANSPORT If you are shipping both the centrifuge and a rotor, remove the rotor from the centrifuge and pack it in its original shipping container. A rotor left in the centrifuge can bend the drive shaft if the package is jarred during shipment Storage Before storing a centrifuge for an extended period remove the rotor. Return the centrifuge to its original shipping container to protect it from dust and dirt. Temperature and humidity conditions for storage should meet the operating environment requirements described under SPECIFICATIONS in Section 1. Storage conditions Air Temperature Relative Humidity Atmospheric pressure In transport packaging -25C to 55C 10 to 75% 70 to 106 kpa Without transport packaging -5C to 45C 10 to 75% 70 to 106 kpa 4 5
